Why is hemp protein called a super plant protein?
Hemp seeds are the seeds of hemp and have very high nutritional value. The shelled seed kernels are also called hemp kernels and have the same origin as medicine and food. The physical and chemical properties, nutritional properties and applications of active seed peptides and hemp seed proteins have also been rarely studied and summarized.
Dr. Chen Haihong, a postdoctoral fellow at the Shenzhen International Graduate School of Tsinghua University, Dr. Wang Yi and Professor Xing Xinhui from the Department of Chemical Engineering of Tsinghua University, etc. summarized the extraction and purification methods, physical and chemical properties, nutritional activity, and application and prospects of hemp seed protein in the food industry in this article. At the same time, it also pointed out the challenges in the development and application of hemp protein.
Extraction and purification of hemp seed protein
Protein can be divided into plant protein, animal protein and microbial protein according to its source, and the source of protein also determines its extraction method to a certain extent. The extraction of plant protein usually includes chemical extraction, physical extraction and enzyme-assisted extraction (Figure 1), while the commonly used extraction methods of hemp seed protein include chemical extraction, physical extraction and enzyme-assisted extraction. As a plant seed with extremely high oil content, the extraction of hemp seed protein usually includes steps such as pressing to remove oil, extraction, concentration and separation. The presence of oil will lead to the formation of lipoprotein complexes, thereby reducing the extraction efficiency, so defatting is considered A necessary step for efficient plant seed protein extraction. Low-temperature pressing and solvent extraction are the most commonly used extraction methods for hemp seed oil. Commonly used organic solvents include petroleum ether, n-hexane, and n-pentane. Differences in the place of origin and degreasing method of hemp seeds will have an impact on the degreasing efficiency of hemp seeds and the protein content in hemp seed meal after pressing. Researchers often use alkali extraction, acid precipitation (isoelectric point precipitation) and micellar extraction methods to separate and purify hemp seed protein. The extraction pH value is generally between 9 and 10, and the pH value of isoelectric precipitation is generally between 3 and 5. Carne et al. used alkali extraction and acid precipitation to separate and purify hemp seed protein and obtain a protein extract with a protein purity higher than 90%. At the same time, the author also compared the differences in protein obtained by acid extraction and alkali precipitation and alkali extraction and acid precipitation. The study found that the purity of the protein isolate obtained by both methods was greater than 90%, and the protein's emulsifying activity, emulsion stability, and amino acid composition All are better than protein powder directly after oil removal. At the same time, it was also found that the protein obtained by alkali extraction and acid precipitation has better water solubility, while the protein obtained by acid extraction has better emulsion stability.

The alkaline acid precipitation method is widely used for the extraction of hemp seed protein. However, the protein powder extracted by micellar technology is whiter in color than the protein obtained by alkali extraction and acid precipitation. In addition, the author also found that there are large differences in the physical and chemical properties of the proteins obtained by the two methods. The isoelectric point of the isolated protein obtained by the alkali extraction and acid precipitation method is 5.0, and the isoelectric point of the isolated protein obtained by the micellar method is 6.0. , and the transition enthalpy of the protein isolate obtained by the alkali extraction and acid precipitation method is higher than that of the protein isolate obtained by the micelle method. In addition, Dapčević-Hadnađev et al. used a micellar salt extraction method to extract hemp seed protein, and the purity of the final protein obtained was 98.9% (dry mass).
